The reaction was performed at pH 6.5 (200 mM, potassium phosphate buffer) and 30 °C with thionine acetate (TA, 20 μM) as the electron acceptor. The initial velocity was monitored by reduction of the electron acceptor at 595 nm using a UV-visible spectrophotometer. The solid line is the fit with the Michaelis-Menten equation (R2 = 0.9972). The experiments were performed in triplicate.
