The cloning method requires only three steps: (A) amplification of insert fragments using primer F1 and R1; (B) exponential amplification using fragments generated in the first PCR and reverse primer R; and (C) digestion by DpnI to eliminate the parent DNA and direct transformation into chemically competent Top10Fâ cells.
