(A) After CMECs were incubated with TNF-α for 30 min with Gö 6983 or Gö 6976 pre-treatment, membrane and cytosol fractions were collected for immunoblotting; *P < 0.05 versus control (Ctr); #P < 0.05 or ##P < 0.01 versus TNF-α (n = 3). (B–F) CMECs were transfected with PKCδ siRNA or PKCζ siRNA for 48 h. Protein expression was analyzed by immunoblotting after TNF-α treatment for 30 min (C) or 18 h (B,D) (n = 3). NF-κB activity was measured with TNF-α treatment for 30 min (E) (n = 4). BODIPY-PA transcytosis was measured after TNF-α treatment for 18 h (F) (n = 4). *P < 0.05 or **P < 0.01 versus Ctr siRNA; #P < 0.05 or ##P < 0.01 versus Ctr siRNA+TNF-α.
