Figure 1

(a) Transmission electron microscopy of BEAS-2B and Stard7KDBEAS-2B cells detected smaller mitochondria and poorly defined cristae in knockdown cells compared to WT cells. NUC: nucleus. (b) Images of mitochondria labeled by TOMM22 antibody were collected from BEAS-2B (n = 47) and Stard7KDBEAS-2B cells (n = 48) for morphometric analyses of surface area (A). Stard7KDBEAS-2B cells had higher than expected numbers of smaller mitochondria (A ≤ 0.5 μm2) compared to BEAS-2B cells (*p = 0.0045). (c) Mitochondrial membrane potential (Δψm) was assessed after incubation with or without DilC1(5) for 30 minutes followed by flow cytometry. Stard7 deficiency was associated with decreased Δψm. BEAS-2B cells vs. Stard7KDBEAS-2B cells, ***p < 0.0001. Transient transfection of Stard7KDBEAS-2B cells with Stard7-1HA partially restored Δψm. Stard7KDBEAS-2B/Stard7-1HA vs. Stard7KDBEAS-2B cells, ***p < 0.0001. MFI, mean fluorescence intensity.
