MASTER-NAADP: a membrane permeable precursor of the Ca²⁺ mobilizing second messenger NAADP

Upon stimulation of receptors, NAADP is formed as calcium-mobilizing second messenger in many cells. Here, the authors develop MASTER-NAADP, a membrane-permeant precursor of NAADP, and characterize its biological activity.

2′-Deoxyadenosine 5′-diphosphoribose is an endogenous TRPM2 superagonist

2′-Deoxy-ADPR, formed from the combined action of NMNAT and CD38 in response to H₂O₂ signaling, activates TRPM2 ion channel currents more potently than the known agonist ADPR via a deceleration of channel inactivation and a higher average open probability.

Short-term dietary changes can result in mucosal and systemic immune depression

Here, the authors show that short-term consumption of energy-dense diets deficient in fiber, similar to eating patterns for many people today, results in a transient depression of the mucosal and systemic immune systems such that susceptibility to bacterial infection is increased.

TGF-β signaling in Th17 cells promotes IL-22 production and colitis-associated colon cancer

Poly-functional helper T cells can have a stronger effect than mono-functional T cells, but whether the response is qualitatively different is not clear. Here the authors show that a population of IL-17⁺IL-22⁺, but not single IL-22⁺, CD4⁺ T cells are induced by TGF-β, enriched in patients with colorectal cancer (CRC) and drive CRC progression in mice.
