Engineering and characterization of gymnosperm sapwood toward enabling the design of water filtration devices

Gymnosperm sapwood is an abundantly available material to construct water filters but the material’s behaviour as a filter is poorly understood and challenges such as short shelf live have not been addressed. Here, the authors develop guidelines for the design and fabrication of xylem filters and demonstrate gravity-operated filters with a shelf life of more than two years for the removal of contaminants from spring, tap and ground water.

DNA2 suppresses recombination-restarted replication and checkpoint activation at stalled forks, and its loss triggers recombination-dependent synthesis, checkpoint signalling and cell-cycle exit, highlighting its essential role in proliferation and growth failure in primordial dwarfism.

Recombination-restarted replication makes inverted chromosome fusions at inverted repeats

A new mechanism of chromosomal rearrangement is identified through the observation that broken or collapsed DNA replication forks restarted by homologous recombination have a high propensity for U-turns at short inverted repeats; the error-prone nature of this mechanism is suggested to contribute to gross chromosomal rearrangements and copy-number variations present in cancer and other genomic disorders.

The authors uncover synthetic lethality between TP53 and ENDOD1, which contains an atypical endonuclease domain. Co-depleted G1 cells accumulate single stranded DNA and die. Combined with xenograft data the work identifies ENDOD1 as a potential cancer-specific SL drug discovery target.

Smc5/6: a link between DNA repair and unidirectional replication?

The structural maintenance of chromosomes (Smc)5/6 complex has a poorly characterized role in DNA repair. Smc5/6 has been implicated specifically in rDNA stability, but the authors propose that the unidirectional replication of rDNA merely accentuates the genome-wide functions of Smc5/6 in repairing DNA replication errors.

Homologous recombination can overcome replication fork inactivation, but this can cause gross chromosomal rearrangements.
