  • Wang et al show that microglial NF-κB activation is essential for tau spreading and tau-mediated spatial learning and memory deficits in tauopathy mice. Inactivation of NF-κB reversed tau associated microglial states and rescued autophagy deficits.

  • Mechanisms of cognitive resilience against Tau pathology are unknown. The authors show that inactivation of the microglial cGAS–interferon axis confers such resilience by preserving the neuronal expression of myocyte enhancer factor 2c.

  • Regulated trafficking of major histocompatibility complex class II and CD86 is a prerequisite of antigen presenting cell functionality. Authors show here that ubiquitin-like protein 3 is critically involved in the ubiquitination process that controls trafficking, with wide-ranging immunological consequences.

  • Disruptions in the autophagy–lysosome pathway in neurons have been implicated in Alzheimer’s disease. A study now reports that autophagy is also critical for disease-associated microglia surrounding amyloid plaques and is protective against microglial senescence and neuropathology in an Alzheimer’s disease mouse model.
