Lacabanne and Orelle et al. investigate a multidrug ABC transporter BmrA mutant defective in the X-loop, which uncouples ATP hydrolysis from substrate translocation. This study suggests that stiffening of a defined set of amino-acid residues allows the pump to transport substrates properly.
