Discrete and conserved inflammatory signatures drive thrombosis in different organs after Salmonella infection

Infection may induce multi-organ thrombosis, but the underlying inflammatory mechanism remains elusive. Here, using Salmonella Typhimurium infectious mouse model, the authors reveal organ specific and broad inflammatory events like TNF for thrombosis in liver and spleen.

VSV^∆M51 drives CD8⁺ T cell-mediated tumour regression through infection of both cancer and non-cancer cells

The role of non-cancer cells infected by oncolytic viruses (OV) in cancer regression remains elusive. Here the authors engineer OV-sensitive and OV-resistant cancer cell lines and show that OV infection of non-cancer cells can elicit effective antitumour immunity via enhancing DC function and CD8⁺ T cell activation.

Denisovan, modern human and mouse TNFAIP3 alleles tune A20 phosphorylation and immunity

A20, encoded by TNFAIP3, is a negative-feedback inhibitor of NF-κB. Grey and colleagues identify natural human variants of TNFAIP3, which lower A20 activity and increase autoinflammatory responses. These alleles were inherited by descendants of Denisovans who crossed the Wallace Line to inhabit Oceania.

Neutrophils and cancer: guilt by association

A new study reports that activation of neutrophils leads to the release of neutrophil extracellular traps (NETs) and coagulation, which then promote development of a tumorigenic microenvironment in the small intestine. These findings provide novel insights into the role of neutrophils in cancer and suggest enticing directions for future research.

Smac mimetics and oncolytic viruses synergize in driving anticancer T-cell responses through complementary mechanisms

Oncolytic viruses (OV) and second mitochondrial activator of caspase (Smac)-mimetic compounds (SMC) synergistically kill cancer cells directly. Here, the authors show that SMC and OV therapies combination also synergize in vivo by promoting anticancer immunity through an increase in CD8⁺ T-cell response.

Infection-induced NETosis is a dynamic process involving neutrophil multitasking in vivo

Bacteria can be trapped by neutrophil extracellular traps (NETs) in vitro, but their relevance in vivo is uncertain, in part because NETs are thought to be released by dying neutrophils, thereby eliminating the other antimicrobial functions of these cells. Paul Kubes and his colleagues report in this issue that NET release need not kill neutrophils and that NETosis can by dynamically imaged in vivo.
