Suppressing recurrence in Sonic Hedgehog subgroup medulloblastoma using the OLIG2 inhibitor CT-179

Previously, OLIG2-expressing stems cells have been identified as having a role in medulloblastoma recurrence. Here, the authors investigate the effects of targeting this OLIG2+ stem cell population in Sonic Hedgehog (SHH) medulloblastoma using CT-179, an OLIG2 inhibitor.

Although the hedgehog (HH) pathway is known to be deregulated in medulloblastoma, inhibitors of the pathway have shown disappointing clinical benefit. Using single-cell sequencing in a mouse model of the disease, the authors show that the response to the HH pathway inhibitor vismodegib is cell-type specific.

Cryptic developmental events determine medulloblastoma radiosensitivity and cellular heterogeneity without altering transcriptomic profile

Malawsky, Weir et al. compare medulloblastomas that form in mice engineered to express SmoM2, a constitutively active component of the Sonic hedgehog signaling pathway, starting in either neural progenitor or stem cells. Medulloblastomas in the two models show similar bulk transcriptomic profiles, but contain different fractions of tumor stem cells and different myeloid populations, and demonstrate different therapeutic responses.
