Multiple roles of H2A.Z in regulating promoter chromatin architecture in human cells

Histone variant H2A.Z has been suggested to contribute to the regulation of promoter accessibility. Here, the authors present high-depth maps of the position and accessibility of H2A.Z-containing nucleosomes for human Pol II promoters and provide evidence that H2A.Z has multiple and distinct roles in regulating gene expression dependent upon its location in a promoter.

A unique H2A histone variant occupies the transcriptional start site of active genes

The histone variant H2A.Bbd inhibits folding of nucleosomal arrays and reverses chromatin-mediated transcriptional repression in vitro. New studies have uncovered the related mouse H2A variant H2A.Lap1 as a novel component of the transcription start site of active genes during specific stages of spermatogenesis, which enables transcriptional activation by unfolding the chromatin locally.

Histone H2A.Z inheritance during the cell cycle and its impact on promoter organization and dynamics

Histone H2A.Z-containing nucleosomes flank the transcription start sites (TSSs) of active mammalian genes. A new study reveals that histone H2A.Z at the TSS is reduced during S phase, which coincides with its gain at centromeric and subtelomeric regions during M phase and mirrors an expansion of the nucleosome-depleted region, which, surprisingly, is unrelated to transcriptional activity.

Chromatin compaction has profound implications for the regulation of transcription, replication and DNA repair. Changes in nucleosome structure and stability — due to the incorporation of variant histones and post-translational modifications of histones — affect chromatin compaction. Chromatin structures are not nearly as uniform as previously assumed, which should be taken into account in the context of specific biological functions.
