Dominant immune tolerance in the intestinal tract imposed by RelB-dependent migratory dendritic cells regulates protective type 2 immunity

Dendritic cells play intricate roles in engaging a range of immune cells. Here, the authors establish a role for the transcription factor RelB in dendritic cells as a molecular rheostat that controls the level of immune tolerance by limiting the number of regulatory T cells.

Nanofluidic proteomic assay for serial analysis of oncoprotein activation in clinical specimens

Here Fan et al. describe a protein analysis platform for the sensitive, nanoscale diagnosis and investigation of clinical specimens, including monitoring the response to targeted therapeutics. The nanofluidic proteomic immunoassay can be used to quantify total and phosphorylated forms of oncoproteins in small tumor samples and has been validated in vivo in mouse tumors and in clinical specimens from blood, surgical biopsies and fine-needle aspirates.

Eosinophils preserve bone homeostasis by inhibiting excessive osteoclast formation and activity via eosinophil peroxidase

Eosinophils are traditional immune effectors involved in tissue homeostasis. In this study, eosinophils emerge as key regulators of bone homeostasis by interacting with osteoclasts, inhibiting their differentiation and pathological bone loss.

Neutrophil breaching of the blood vessel pericyte layer during diapedesis requires mast cell-derived IL-17A

The blood vessel wall is a complex multi-layered structure, yet upon injury or infection, neutrophil leukocytes are rapidly migrating from the blood stream to the affected tissues, by a process termed diapedesis. Authors here show that the final steps of diapedesis through the outer pericyte layer is regulated by perivascular mast cells via IL-17A production.

Type 2 immune responses are viewed as opposites of Type 1 and 17 responses. Here the authors show that activation of Type 2 immunity by helminth infection counteracts the development of inflammatory arthritis, a type 17-mediated pathology, via IL-4/IL-13- STAT6 signalling and eosinophil activation.

An efficient and versatile system for acute and chronic modulation of renal tubular function in transgenic mice

Traykova-Brauch et al. have developed a new approach to modeling renal diseases such as polycystic kidney disease, renal fibrosis and renal cancer in transgenic mice. In contrast to currently available tools, Pax8-rtTA–transgenic mice have high levels of transgene expression in a highly kidney-specific, uniform and tetracycline–dependent manner. The usefulness of the Pax8–rtTA system, which is both inducible and reversible, has been shown in three different settings.

Chitin induces accumulation in tissue of innate immune cells associated with allergy

The biopolymer chitin is one of the main components in the cell walls of fungi, the exoskeletons of insects and crustaceans and the bodies of some parasitic worms. This paper describes how chitin elicits an innate allergic response in mice, which is controlled by a host chitinase.
