TRIM52 maintains cellular fitness and is under tight proteolytic control by multiple giant E3 ligases

Despite not conserved in all mammals, TRIM52 is crucial for fitness in human cells. Here, the authors show that TRIM52 interacts with the DNA repair machinery to prevent cell-cycle arrest, and that TRIM52 is rapidly degraded by multiple giant E3 ligases. These findings link TRIM52 to genome integrity control.

Chemical inhibition of RNA viruses reveals REDD1 as a host defense factor

A screen for compounds that alleviate the inhibitory effect of influenza NS1 on host gene expression and suppress viral toxicity found naphthalimides that could upregulate REDD1, an mTORC1 inhibitor, revealing that viruses inhibit REDD1 to activate the mTORC1 pathway.

Negative regulation of NF-κB activity by brain-specific TRIpartite Motif protein 9

ß-TrCP is an adaptor protein that controls activity of several key regulatory proteins including NFκB by ubiquitin-mediated proteolysis. Here Shi et al. demonstrate that ß-TrCP is negatively regulated by the brain-specific protein TRIM9, limiting activation of NFκB and production of proinflammatory cytokines.
