A single three-dimensional chromatin compartment in amphioxus indicates a stepwise evolution of vertebrate Hox bimodal regulation

José Luis Gómez-Skarmeta, Hector Escrivá, Ignacio Maeso, Damien Devos and colleagues perform 4C-seq profiling of the Hox cluster in amphioxus embryos and find that, unlike in vertebrate embryos, the cluster is organized into a single chromatin interaction domain. They suggest that the vertebrate Hox bipartite regulatory system is an evolutionary novelty.

Nodal–Activin pathway is a conserved neural induction signal in chordates

Little is known about the evolution of neural induction mechanisms in chordates. Here, the authors show that the dorsal blastopore lip of the cephalochordate amphioxus is homologous to the vertebrate dorsal organizer and that it promotes neural induction through Nodal–Activin signal.

The emergence of the brain non-CpG methylation system in vertebrates

By studying brain DNA methylation across 13 distantly related animals, the authors show that non-CpG DNA methylation, which plays a regulatory role in cognition, is restricted to vertebrates and was assembled at the origin of the vertebrate lineage as a result of the ancestral vertebrate whole-genome duplication.
