NKp46 enhances type 1 innate lymphoid cell proliferation and function and anti-acute myeloid leukemia activity

It has been previously reported that the natural cytotoxic receptor NKp46 plays an important role in regulating natural killer (NK) cell biology. Here the authors show that NKp46 is required for the proliferation and cytotoxicity of type 1 innate lymphoid cells (ILCs) by activating the NF-κB signaling pathway and regulating IL-2Rα expression.

Synthesis of nimbolide and its analogues and their application as poly(ADP-ribose) polymerase-1 trapping inducers

Nimbolide is an inhibitor of poly(ADP)-ribosylation-dependent ubiquitin E3 ligase RNF114. A modular synthesis of nimbolide and its analogues is now reported, using a sulfonyl hydrazone-mediated etherification and a radical cyclization strategy. These analogues give insight into structure–activity relationships and some have superior poly(ADP-ribose) polymerase-1 trapping activity relative to nimbolide.

The intrinsically disordered protein TgIST from Toxoplasma gondii inhibits STAT1 signaling by blocking cofactor recruitment

Intrinsically disordered proteins (IDP) are pleotropic proteins with diverse functions. Here the authors show that an IDP, TgIST, from T. gondii blocks interferon-induced gene expression by binding to the STAT1 dimer interface and preventing the recruitment of co-transcriptional activators, CBP/p300, to STAT1 to inhibit expression of immunity genes.
