Self-amplifying NRF2–EZH2 epigenetic loop converts KRAS-initiated progenitors to invasive pancreatic cancer

Using pancreatic organoids and genetic mouse models, Antonucci et al. show that cellular stress activates an NRF2–EZH2 epigenetic axis leading to metabolic reprogramming and subsequent malignant transformation of KRAS-mutant benign lesions.

Mitochondrial DNA oxidation propagates autoimmunity by enabling plasmacytoid dendritic cells to induce T_FH differentiation

Oxidized mitochondrial DNA can be released into cytoplasm activating the Nlrp3 inflammasome resulting in its extracellular release. Here the authors show that only this oxidized form can induce autoantibody production by uptake into plasmacytoid dendritic cells, which then produce interleukin-21 to differentiate naive T cells into T_FH cells.

Mitophagy plays a critical role in cellular homeostasis, and PINK1/Parkin-mediated mitophagy is the most thoroughly characterized. Here, Xian et al. show that STX17 induces mitophagy via a macroautophagy pathway regulated by Fis1, by a PINK1/Parkin-independent route.
