Sulfur-containing class of broad-spectrum antivirals improves influenza virus vaccine development

Improved vaccines and antivirals are needed for many enveloped viruses. Here, the authors identify sulfur-based small molecules that disrupt viral membrane properties, inhibiting fusion and entry, and safely inactivate influenza virus. The resulting inactivated influenza vaccine is protective in mice.

The reduction of nitrite (NO₂⁻) to nitric oxide (NO), relevant to the biogeochemical nitrogen cycle as well as radioactive waste, typically occurs at redox-active metal centres. Now, a Lewis acid-capped nitrite has been reduced to the nitrite dianion (NO₂²⁻), a nitrogen-centred radical that connects three redox levels in the global nitrogen cycle through NO₂⁻, NO and N₂O.

Diphthamide biosynthesis requires an organic radical generated by an iron–sulphur enzyme

Translation elongation factor 2 (EF2) from archaea and eukaryotes contains a unique, post-translationally modified histidine residue called diphthamide, which is the target of diphtheria toxin. The biosynthesis of diphthamide involves three steps; here it is shown that the first step in the archaeon Pyrococcus horikoshii requires an unusual iron–sulphur-cluster enzyme, Dph2. It catalyses unprecedented chemistry.

Conformational ensemble of the sodium-coupled aspartate transporter

The sodium and aspartate symporter Glt_Ph mediates transport by alternating between outward-facing and inward-facing states. These conformational changes are now probed using double electron-electron resonance (DEER) spectroscopy. The data show that Glt_Ph samples both states with similar probabilities, and that each protomer in the Glt_Ph homotrimer behaves independently of the others.

Expanded intronic G₄C₂ repeats in C9orf72 cause ALS/FTD. GR dipeptide repeats produced in C9orf72 ALS/FTD co-aggregate with Kap2 in neurons, affecting survival. Upregulating Kapβ2 may mitigate neurotoxicity, a potential therapy for C9orf72-ALS/FTD.
