The ground state of embryonic stem cell self-renewal

Smith and colleagues show that embryonic stem cell self-renewal does not rely on extrinsic instruction, and can be enabled by the elimination of factors that induce differentiation and by inhibition of glycogen synthase kinase 3. They interpret the data as showing the ground state of the embryonic stem cell in the absence of extrinsic instruction is self renewal.

GSK-3 is a master regulator of neural progenitor homeostasis

The authors report the conditional deletion of the alpha and beta forms of glycogen synthase kinase 3 (GSK-3) in mouse neural progenitors. This deletion is associated with dysregulations in β-catenin, Sonic Hedgehog, Notch and fibroblast growth factor signaling pathways and leads to markedly increased proliferation of neural progenitors.

Gsk3 is a metabolic checkpoint regulator in B cells

Mature B cells remain in a quiescent state until activated. Rickert and colleagues identify a prominent role for the kinase Gsk3 in resting naive B cells and in activated germinal center B cells that restrains the production of Myc and reactive oxygen species and prevents metabolic collapse.
