SV2A, an essential membrane protein in neurotransmission, is known as the receptor of clostridial neurotoxins and target of anti-epileptic drugs. Here the authors show details of the binding site for the drug Levetiracetam and the recognition of Tetanus Neurotoxin.

Global kinome profiling reveals DYRK1A as critical activator of the human mitochondrial import machinery

Mitochondrial protein import is mediated by the translocase of the outer membrane (TOM), through which nearly all precursors traverse. Here, the authors perform global in vitro kinome profiling and by this identify that DYRK1A phosphorylates TOM70 and promotes import.

Beware of extreme calculated lipophilicity when designing cyclic peptides

Orally bioavailable, high molecular weight macrocyclic peptides that inhibit difficult-to-drug protein–protein interactions are of high therapeutic value, and rules for their design were proposed recently. Here, we emphasize the danger of rules that provide a false impression of the lipophilicity required of a clinical candidate.

Expulsion mechanism of the substrate-translocating subunit in ECF transporters

The final step in an ECF transporters transport cycle involves the expulsion of the membrane embedded substrate binding protein (the S-component) from the motor (the ECF module). Here the authors show how the motor uses ATP binding to load a molecular spring, and adjusts the shape of the membrane, to achieve this step.

Expanding functional protein sequence spaces using generative adversarial networks

A protein’s three-dimensional structure and properties are defined by its amino-acid sequence, but mapping protein sequence to protein function is a computationally highly intensive task. A new generative adversarial network approach learns from natural protein sequences and generates new, diverse protein sequence variations, which are experimentally tested.

Reiner Gamma albedo features reproduced by modeling solar wind standoff

Since the Renaissance, the Reiner Gamma formation was identified as a peculiar lunar feature. By combining magnetic field topology based on satellite observations with a full-kinetic plasma model, the authors successfully reproduced the shape of the surface albedo pattern beneath the magnetic anomaly.
