K63-linked polyubiquitination of transcription factor IRF1 is essential for IL-1-induced production of chemokines CXCL10 and CCL5

IL-1 triggers IRF1 signaling, but the biological importance of this has remained uncertain. Kordula and colleagues show that an IL-1–IRF1 pathway is necessary for the selective induction of chemokines and sterile inflammation.

Active, phosphorylated fingolimod inhibits histone deacetylases and facilitates fear extinction memory

Fingolimod is a sphingosine-1 phosphate (S1P) receptor modulator and an immune modulator in use as a treatment for the remitting-relapsing form of multiple sclerosis. Hait et al. show that the active form of fingolimod can inhibit neuronal class I histone deacetylases (HDACs), modulate gene expression in the brain and facilitate memory extinction. The authors also show spatial and associative memory deficits in mutant mice lacking the enzyme necessary for formation of the active form of fingolimod.

Sphingosine-1-phosphate is a missing cofactor for the E3 ubiquitin ligase TRAF2

Engagement of the tumour-necrosis factor (TNF) receptor results in the assembly of multi-component signalling complexes by adaptor proteins that include TNF receptor-associated factor 2 (TRAF2). Genetic evidence indicates that TRAF2 is needed for the polyubiquitination of receptor interacting protein 1 (RIP1), but direct evidence has been lacking. Here it is shown that the lipid sphingosine-1-phosphate is a co-factor needed for this ubiquitination activity of TRAF2.
