The value of political ecology in biodiversity conservation

Political ecology aims to understand how politics and power influence both social and ecological dynamics. Conservation biologists and political ecologists tackle many of the same pressing environmental problems, but from quite different perspectives. In this Viewpoint, five political ecologists discuss their views on how the field can understand and tackle the biodiversity crisis.

Approaches to financing biodiversity conservation tend to focus on funding gaps, but fail to address underlying political and economic drivers. We propose two strategies — tax reform and debt justice — to supercharge public financing for biodiversity and deflate harmful financial flows, while chipping away at the causes of state austerity.

Hallmarks of CD8⁺ T cell dysfunction are established within hours of tumor antigen encounter before cell division

Rudloff et al. examine the kinetics of CD8⁺ T cell dysfunction/exhaustion. Tumor-specific CD8⁺ T cells in the tumor environment exhibit epigenetic modifications within hours, before cell division. The findings suggest a temporal relationship between tumor antigen exposure, chromatin remodeling and dysfunction ‘imprinting’.

A latent subset of human hematopoietic stem cells resists regenerative stress to preserve stemness

Dick and colleagues identify human LT-HSC subsets with distinct quiescent states. They link these differences to INKA1-mediated downregulation of the transmembrane protein CD112 and its interaction with the protein deacetylase SIRT1. INKA1 is inversely correlated with the histone H4K16Ac mark, which then distinguishes ‘latent’ CD112^lo LT-HSCs from CD112^hi LT-HSCs that are more readily activated in response to hematopoietic stress.

Thomas and colleagues examine preinfection baseline parameters of cellular and serologic immunity. Their findings collectively show that peripheral cell composition provides better correlates of immune protection from symptomatic influenza infection than vaccination, demographics or serology alone.

The multiple myeloma microenvironment is defined by an inflammatory stromal cell landscape

Multiple myeloma disease progression and therapy response are influenced by the bone marrow niche in which the tumor cells reside. To characterize this supportive niche, Cupedo and colleagues use single-cell transcriptomic analysis of bone marrow stromal cell populations from individuals with multiple myeloma. They identify a myeloma-specific inflammatory mesenchymal stromal cell (iMSC) population that spatially colocalizes with tumor cells. Anti-myeloma induction therapy does not influence iMSC presence, suggesting a role for bone marrow inflammation in myeloma persistence or relapse.
