ATP induces folding of ALS-causing C71G-hPFN1 and nascent hSOD1

Amyotrophic lateral sclerosis (ALS) is associated with the misfolding of hSOD1 and C71G-hPFN1, however, the exact mechanism responsible for ALS remains poorly understood. Here, NMR spectroscopy reveals that ATP can induce the folding of hSOD1 and C71G-hPFN1, inhibiting protein aggregation and enhancing their thermodynamic stability.

ATP biphasically modulates LLPS of TDP-43 PLD by specifically binding arginine residues

Dang Mei et al. use NMR and microscopy approaches to examine how ATP impacts the liquid-liquid phase separation (LLPS) of prion-like domains in TDP-43, a RNA-binding protein that is implicated in ALS and other neurological disorders. Their results suggest that ATP specifically binds to a subset of TDP-43 arginine residues at a particular molar ratio to modulate LLPS, and provides insight into how ATP affects the LLPS of biomolecular systems.

ATP binds and inhibits the neurodegeneration-associated fibrillization of the FUS RRM domain

Kang et al investigate by NMR and modeling methods how ATP influences the neurodegeneration–associated protein FUS. They find that ATP binds the RNA-binding RRM domain of FUS and inhibits its fibrillization without affecting ligand binding or thermodynamic stability.
