The mechanism of YAP/TAZ transactivation and dual targeting for cancer therapy

The mechanism underlying YAP/TAZ transactivation remains to be explored. Here the authors report that the hydrophobic transcriptional activation domains of YAP/TAZ exert dual functions on transcriptional initiation and elongation through interactions with TAF4 and MED15, which could be targeted for anti-cancer therapy.

LATS2 condensates organize signalosomes for Hippo pathway signal transduction

Qin et al. find that cell detachment induces condensation of LATS2, a core kinase of the Hippo pathway. These LATS2 condensates protect LATS2 from degradation by the ubiquitin–proteasome system, thereby promoting the assembly of Hippo signalosomes for pathway activation.
