Energy-dependent regulation of cell structure by AMP-activated protein kinase

AMP-activated protein kinase (AMPK) is important for mitosis and epithelial cell polarity and these effects are a result of direct phosphorylation of non-muscle myosin regulatory light chain by AMPK. This is the first study reporting AMPK deletion in an entire organism and highlights the physiological function of this kinase.

cAMP signalling in mushroom bodies modulates temperature preference behaviour in Drosophila

This study shows that mushroom bodies in Drosophila brains also have a role in temperature preference behaviour. When cAMP-dependent kinase activity in mushroom body neurons is artificially lowered, the Drosophila cannot find their desired temperature properly. When it is increased, they prefer higher temperatures. The study adds a classic homeostatic process to the mushroom bodies' list of functions.

Lysosomal control of senescence and inflammation through cholesterol partitioning

Senescenct cells are shown to accumulate cholesterol in lysosomes, which upregulates mTORC1 signaling, thereby supporting the senescence-associated secretory phenotype and promoting senescence-associated inflammation.

A metazoan ortholog of SpoT hydrolyzes ppGpp and functions in starvation responses

SpoT has a key role in the bacterial starvation response. Now the metazoan ortholog of SpoT, Mesh1, has been identified, and the structure reveals a conserved active site that can catalyze ppGpp hydrolysis. The Drosophila Mesh1 deletion mutant has impaired starvation resistance, and microarray analysis gives further insight into the starvation response.

Diabetic sensory neuropathy and insulin resistance are induced by loss of UCHL1 in Drosophila

The mechanisms underlying diabetic neuropathy remain elusive. Here, the authors identify that UCHL1 deubiquitinase positively regulates insulin signaling and its loss leads to axonal degeneration of sensory neurons.

PINK1 and Parkin regulate IP₃R-mediated ER calcium release

Loss of two PD genes, PINK1 and Parkin, leads to a robust increase in ER calcium release. Here, the authors show that suppression of IP₃R activity via inhibiting CISD1 is sufficient to rescue the PD-related phenotypes in PINK1 or Parkin null animal models.
