ATM orchestrates the DNA-damage response to counter toxic non-homologous end-joining at broken replication forks

Mutations in the ATM tumor suppressor gene confer hypersensitivity to DNA-damaging chemotherapeutic agents. Here, the authors provide evidence that these hypersensitivities reflect a crucial role for ATM at damaged replication forks being to prevent toxic DNA end-joining leading to chromosome fusions and cell death.

H2AX promotes replication fork degradation and chemosensitivity in BRCA-deficient tumours

Histone H2AX has a known role in DNA damage repair but interestingly, its loss is associated with resistance to poly(ADP-ribose) polymerase (PARP) inhibition in BRCA-deficient tumours. Here, the authors identify a role of γH2AX in the degradation of replication forks and demonstrate that H2AX loss drives PARP inhibitor resistance via increased stressed fork stability in BRCA-deficient tumours.

CtIP tetramer assembly is required for DNA-end resection and repair

CtIP helps maintain genomic stability by promoting DNA double-strand-break repair. Structural and biophysical analyses now show that the N terminus of human CtIP forms a tetrameric structure that is required for resection of broken DNA ends to permit their repair by homologous recombination.

Spin-crossover nanoparticles anchored on MoS₂ layers for heterostructures with tunable strain driven by thermal or light-induced spin switching

Spin-crossover nanoparticles have been covalently grafted onto a semiconducting MoS₂ layer to form a self-strainable heterostructure. Under light or thermal stimulus, the nanoparticles switch between their high- and low-spin states, in which they have different volumes. This generates a reversible strain over the MoS₂ layer and, in turn, alters the electrical and optical properties of the heterostructure.

Here the authors identify that C16orf72 regulates BRCA1/Senataxin to promote replication fork recovery. These proteins act together in a pathway parallel to PARP1 to suppress R-loop accumulation in response to replication stress and confer resistance to PARP inhibitors.

Small-molecule–induced DNA damage identifies alternative DNA structures in human genes

Identifying DNA sequences that adopt alternative structures within the context of genomic DNA presents a major challenge. Pyridostatin, a G-quadruplex–specific chemical probe, was shown to induce DNA damage at specific genomic sites, including the proto-oncogene SRC, leading to cell cycle arrest in human cancer cells.
