The editing domains of AlaRS (AlaRS-Ed) and ThrRS (ThrRS-Ed) are homologous, yet they differ in sensitivity to reactive oxygen species (ROS). Here, the authors reveal the mechanistic basis of how the universally conserved Zn2+ ion safeguards AlaRS-Ed’s activity from ROS, while its absence in ThrRS-Ed renders its hypersensitive.
