Single cell sequencing identifies clonally expanded synovial CD4⁺ T_PH cells expressing GPR56 in rheumatoid arthritis

Antibodies against citrullinated proteins (ACPA) can divide rheumatoid arthritis patients but the difference in immune phenotype is not clear. Here the authors characterise T peripheral helper cells in arthritic joints comparing ACPA+ and ACPA- patients showing changes in immune cell phenotype and surface molecules.

Citrulline immunity in RA: CD8⁺ T cells enter the scene

The involvement of citrulline-specific CD4⁺ T cells in anti-citrulline protein antibody-positive rheumatoid arthritis (RA) is well described, whereas less attention has been given to CD8⁺ T cells. New data suggest that CD8⁺ T cells also contribute to citrulline-specific immune responses in RA.

Recessive mutations in POLR1C cause a leukodystrophy by impairing biogenesis of RNA polymerase III

Some RNA polymerase (POLR) 3-related leukodystrophy cases do not have the causal mutations in POLR3A and POLR3B. Here, by exome sequencing, the authors identify recessive mutations in POLR1C, a gene encoding a shared POLR1 and POLR3 subunit, impairing assembly and nuclear import of POLR3, but not POLR1.

VAMP7 controls T cell activation by regulating the recruitment and phosphorylation of vesicular Lat at TCR-activation sites

How the adaptor Lat and the TCR come together after TCR triggering is not well understood. Hivroz and colleagues show that the vesicular protein VAMP7 is required for the recruitment of Lat-containing vesicles to TCR-activation sites.
