Transcription factors mediate condensin recruitment and global chromosomal organization in fission yeast

Ken-ichi Noma and colleagues use ChIA-PET to identify genome-wide associations mediated by condensin and cohesin in fission yeast. They find that cohesin and condensin generate small and larger chromatin domains, respectively, and that condensin, but not cohesin, connects cell cycle–regulated genes bound by mitotic transcription factors.

Here, the authors report that METTL3 orchestrates cellular senescence by coordinating its enzymatic activity-dependent and independent functions. METTL3-mediated chromatin loops induce phase separation via metabolic reprogramming.

The human Pol III transcriptome and gene information flow

In this issue, three papers report the distribution of the RNA polymerase III (Pol III) machinery, including transcription factor IIIB, transcription factor IIIC and Pol III itself, across the human genome. These studies reveal cell type–specific expression of Pol III genes, functional interplay between the Pol II and Pol III transcriptional machineries and the potential involvement of Pol III genes in chromosome organization.

Involvement of condensin in cellular senescence through gene regulation and compartmental reorganization

Cell senescence involves stable arrest of cell proliferation and changes in gene expression and 3D genome reorganization. Here, the authors show that human condensin II complex participates in reorganization of the chromatin compartments, primarily through switching from heterochromatic B to euchromatic A compartments.

Architectural alterations of the fission yeast genome during the cell cycle

Analysis of the 3D genomic organization of Schizosaccharomyces pombe during the cell cycle reveals that condensin mediates formation of large domains that serve as chromosomal compaction units, whereas cohesin forms smaller, more stable domains.

Epigenetic specifications of host chromosome docking sites for latent Epstein-Barr virus

Epstein-Barr virus (EBV) episomes tether to the host chromosome via EBNA1. Here, using circular chromosome conformation capture (4C), Kim et al. identify attachment sites and show that EBV episomes preferentially associate with transcriptionally silenced genes in Burkitt lymphoma cells.

NAD⁺ metabolism governs the proinflammatory senescence-associated secretome

Nacarelli et al. show that the nicotinamide-phosphoribosyltransferase-regulated NAD⁺ biogenesis pathway promotes the proinflammatory senescence-associated secretory phenotype by enhancing glycolysis and mitochondrial respiration during senescence.

Host genome surveillance for retrotransposons by transposon-derived proteins

A transposon surveillance and genome organization mechanism is described in fission yeast. CENP-B proteins bind to Tf2 retrotransposons and mediate their silencing by recruiting histone deacetylases. CENP-B proteins are also required for the clustering of Tf2 retrotransposons into network-like structures in the nucleus.
