Distinct loops in arrestin differentially regulate ligand binding within the GPCR opsin

Following retinalcis/trans isomerisation, the active form of the G-protein-coupled receptor rhodopsin decays to opsin and all-trans-retinal. In this study, arrestin, a regulator of G-protein-coupled receptor activity, is shown to facilitate the concurrent sequestering of toxic all-trans-retinal and regeneration of 11-cis-retinal within the opsin population.

Strontium isoscape of sub-Saharan Africa allows tracing origins of victims of the transatlantic slave trade

Strontium isotope analysis can be applied to animal and plant tissues to help determine their provenance. Here, the authors generate a strontium isoscape of sub-Saharan Africa using data from 2266 environmental samples and demonstrate its efficacy by tracing the African roots of individuals from historic slavery contexts.

Crystal structure of opsin in its G-protein-interacting conformation

Rhodopsin is a light-activated G-protein-coupled receptor. The structure of its ligand-free state at 2.9 Å resolution was reported recently. This paper describes a similar resolution crystal structure of the same protein bound to a fragment of G-protein and suggests a mechanism by which G-proteins might be activated by G-protein-coupled receptors.

A ligand-specific blockade of the integrin Mac-1 selectively targets pathologic inflammation while maintaining protective host-defense

Integrin-based therapeutics could block inflammatory processes but they also impair host defence, limiting their usefulness. Here the authors report an anti-Mac1 antibody that blocks its interaction with pro-inflammatory ligand CD40L but not other ligands, and show that it can protect against sepsis in mice.

It remains unclear when and why the world’s oceans, once largely occupied by bacteria, became dominated by photosynthetic algae. Polo-like kinase 1 is a key regulator of mitosis and is a candidate for drug development to treat cancer. Here, reduced expression of polo-like kinase 1 in adult mice has a minor impact on animal physiology, suggesting that polo-like kinase 1 inhibitors may be useful in the killing of tumour cells while sparing normal cells.

Recent genetic connectivity and clinal variation in chimpanzees

Lester and colleagues use faecal samples and genetic analyses to investigate the genetic connectivity across chimpanzees. Their results indicate that the global pattern of genetic diversity in chimpanzees is largely characterized by a pattern of isolation by distance with several isolated populations exhibiting strong local differentiation.
