Niu, Shen, Huang et al. presents SAFE Hi-C, a Hi-C library preparation method that uses ligation products to prepare a library for deep sequencing without amplification. They show that SAFE Hi-C can build a chromatin interaction network more faithfully than traditional Hi-C by preserving higher complexity of chromatin interaction and lowering sequencing depth.
