  • An X-ray crystal structure of sarcoplasmic reticulum Ca2+-ATPase (SERCA) in the presence of sarcolipin, a SERCA regulator, is presented; the structure shows that sarcolipin traps SERCA in a previously unidentified ‘open’ state in which its high-affinity Ca2+-binding sites are unoccupied, but accessible from the cytoplasm.

  • The Na+/K+-ATPase pumps three sodium ions out of and two potassium ions into the cell while splitting a single molecule of ATP. Here it is found that the carboxy terminus of the ATPase's α-subunit is also a key regulator of a previously unrecognized ion pathway. The data indicate that, in the ATPase's potassium-bound state, a cytoplasmic proton can enter and stabilize site III when empty. When potassium is released, the proton returns to the cytoplasm, thus permitting an overall asymmetric stoichiometry of the transported ions.

  • Ramachandran plots are a convenient means of describing protein backbone conformation by depicting the distribution of Ca bond rotations. Here, the authors devise an alternative descriptor based on hydrogen bond rotations, and apply it to describe protein structures using a vocabulary of 30 hydrogen-bonding motifs.
