Lineage-restricted function of the pluripotency factor NANOG in stratified epithelia

The transcription factor Nanog regulates self-renewal in pluripotent stem cells and cancer stem cells. Here the authors show that Nanog is expressed in mouse adult stratified epithelia, and its overexpression increases proliferation and aneuploidy and activates pathways associated to mitosis.

Reprogramming in vivo produces teratomas and iPS cells with totipotency features

Induced pluripotent stem cells (iPS cells) have been created in vivo by reprogramming mouse somatic cells with Oct4, Sox2, Klf4 and c-Myc; these cells have totipotent features that are missing from in vitro created iPS cells or embryonic stem cells.

The APC/C cofactor Cdh1 prevents replicative stress and p53-dependent cell death in neural progenitors

The E3-ubiquitin ligase APC/C and its cofactor Cdh1 have been shown to play important roles in axonal growth and synaptic plasticity. In this study, Eguren et al.show that elimination of Cdh1 in the developing nervous system results in defects in the neural progenitor compartment, hydrocephalus and reduced lifespan.

What causes hematopoietic stem cell loss of functionality? Here, Alvarez et al. show that loss of origin licensing factor MCM3 induces replicative stress (RS), causing aberrant erythrocyte maturation, but mice strains with higher tolerance to RS can overcome this defect.

A mouse model of ATR-Seckel shows embryonic replicative stress and accelerated aging

Oscar Fernandez-Capetillo and colleagues report a mouse model of the human Seckel syndrome characterized by a deficiency in ATR. The Seckel mice show high levels of replicative stress during embryogenesis, and the adults show premature aging.

Plk1 regulates contraction of postmitotic smooth muscle cells and is required for vascular homeostasis

The kinase Plk1 has been studied primarily as a mitotic regulator in dividing cells, but de Cárcer et al. find that Plk1 deficiency or inhibition in mice causes nonmitotic defects in the vasculature, including aortic aneurysm and rupture, as well as defective vascular smooth muscle contractility. These results recommend a note of caution in the clinical use of PLK1 inhibitors as anticancer agents.

The Ink4/Arf locus is a barrier for iPS cell reprogramming

The Ink4/Arf tumour suppressor locus encodes the three potent tumour suppressors p16^Ink4a, p15^Ink4b and p19^Arf. Here the locus is shown to be rate-limiting for reprogramming, and its transient inhibition significantly improves the generation of induced pluripotent stem cells. Furthermore, ageing is shown to upregulate the Ink4/Arf locus, with less efficient reprogramming seen in cells from old organisms.
