Palmitoylation prevents B7-H4 lysosomal degradation sustaining tumor immune evasion

The immune checkpoint B7-H4 is regulated through post-translational modifications. Here, the authors identify that in breast cancer cells ZDHHC3-catalyzed B7-H4 palmitoylation prevents its lysosomal degradation and maintains immune suppression, which can be targeted by Abemaciclib to enhance lysosome activation independently of CDK4/6 inhibition.

Cancer stem cells: nature versus nurture

Although all cells within a colon cancer may harbour adenomatous polyposis coli (APC) or β-catenin mutations, activation of Wnt signalling is limited to a subpopulation of cells that display cancer stem cell properties. This activation requires a co-stimulatory signal mediated by hepatocyte growth factor, which is produced by tumour-associated myofibroblasts.

Targeting self-renewal, an Achilles' heel of cancer stem cells

Many tumors display a hierarchical organization that is maintained by a self-renewing 'cancer stem cell' population. A new study in mice shows that targeting the self-renewal regulator BMI-1 abrogates the tumorigenic capacity of colon cancer stem cells, providing a new therapeutic strategy (pages 29–36).

Primary tumor-induced immunity eradicates disseminated tumor cells in syngeneic mouse model

Dissemination of tumor cells from the primary site is an early event. Here, the authors show that the early disseminated tumor cells are actively cleared by the host cytotoxic T lymphocytes induced by the primary tumor and that infiltration of granulocytic myeloid-derived suppressor cells counteracts such immune protection and allow metastasis development.

A metallofullerenol nanomaterial, Gd@C₈₂(OH)₂₂, was shown to inhibit growth of several solid cancers in preclinical models and yet exhibit low toxicity. Herein the authors show that Gd@C₈₂(OH)₂₂functions as an inhibitor of breast cancer stem cell function via blocking TGF-β and HIF-1α signalling, while sparing normal tissue.
