ETMR stem-like state and chemo-resistance are supported by perivascular cells at single-cell resolution

Embryonal tumour with multilayered rosettes (ETMR) is a rare and aggressive paediatric brain tumour. Here, the authors analyse intratumour heterogeneity and the tumour microenvironment in ETMR using single-cell and spatial transcriptomics, in vitro cultures, and a 3D forebrain organoid model, finding important aspects – such as the communication with pericytes – for ETMR development and response to therapy.

Miz1 is a binding partner of the transcription factor c-Myc and a regulator of cell cycle progression. Wolf et al. show that inactivation of Miz1 in the mouse central nervous system results in neurodegeneration, and find that Miz1 is essential for the transcriptional regulation of autophagic flux.

The interaction of DNA repair factors ASCC2 and ASCC3 is affected by somatic cancer mutations

The DNA helicase ASCC3 is the largest subunit of the activating signal co-integrator complex (ASCC), and its DNA unwinding activity is required for the AlkBH3/ASCC-dependent DNA de-alkylation repair pathway. Here, the authors identify a minimal stable complex of the two ASCC subunits ASCC2 and ASCC3, determine the complex crystal structure and further show that cancer-related mutations at the interface between both proteins reduce ASCC2–ASCC3 affinity.

Syk kinase signalling couples to the Nlrp3 inflammasome for anti-fungal host defence

Interleukin-1β (IL-1β) is a key pro-inflammatory factor in innate antifungal immunity, but the mechanism by which the mammalian immune system regulates IL-1β production after fungal recognition is unclear. Here it is demonstrated that the tyrosine kinase Syk controls both pro-IL-1β synthesis and Nlrp3 inflammasome activation after cell stimulation with Candida albicans.

Structural basis for diverse N-glycan recognition by HIV-1–neutralizing V1–V2–directed antibody PG16

Some broadly neutralizing antibodies against HIV-1 recognize glycopeptide epitopes in the gp120 V1–V2 region. Now X-ray crystallography and NMR approaches, together with functional analyses of two such antibodies (PG9 and PG16), reveal how glycan binding specificity is achieved. The authors also created a chimeric Fab that showed improved neutralization activity.

Didriksen, Nawaz, et al. identify three novel genetic risk variants for restless legs syndrome and confirm 19 out of 20 previously reported variants through a genome-wide association meta-analysis including nearly half a million individuals. Using expression QTL analysis, they also find that a subset of these loci may have a causal effect on nearby gene expression.
