Nobuo Horikoshi et al. report the use of CRISPR/Cas9 to insert cleavage and repair sites in gene-rich and gene-poor regions in human cell lines. They show that the frequency of double-stranded break repair by homologous recombination is higher in gene-rich regions with high levels of H4K16ac marks.
