  • The transport of L-lactate across plasma membranes is catalyzed by proton-driven monocarboxylate transporters (MCTs) of the SLC16 solute carrier family. Here, the authors present the crystal structures of a bacterial SLC16 homologue with the bound substrate L-lactate and ligand thiosalicylate both in an outward-open conformation and discuss the L-lactate transport mechanism.

  • The L-lactate-specific transporter SfMCT is a model SLC16 family homologue for the determination of inhibitors and potential drugs. Here, starting from a weakly-binding non-transported ligand, nanomolar affinity inhibitors of SfMCT are developed and crystal structures of the transporter-ligand complexes are presented unveiling interactions at the atomic level.

  • Johannes Thoma et al. overexpress outer membrane proteins (Omps) in Escherichia coli and collect the expelled outer membrane vesicles (OMVs) to study Omp assembly, folding and structure. They find that Omps in OMVs show different unfolding pathways compared to Omps reconstituted in artificial lipid membranes.
