A20 is a regulator of necroptosis

The anti-inflammatory molecule A20 inhibits necroptotic cell death by inhibiting ubiquitination of the kinase RIPK3 at the Lys5 residue and preventing excessive formation of the RIPK1-RIPK3 necroptotic complex.

CD4 expression in effector T cells depends on DNA demethylation over a developmentally established stimulus-responsive element

Epigenetic states that are established during thymic T cell development influence functionality of mature T cells. Authors here show that early developmental programming of the Cd4 locus is comprised of DNA-demethylation at a specific stimulus-responsive element, which allows long-term maintenance of activator histone H3K4 trimethylation and transcription.

Activation of the NLRP1b inflammasome independently of ASC-mediated caspase-1 autoproteolysis and speck formation

The NLRP1b inflammasome activation may lead to pyroptosis and secretion of the inflammatory cytokines IL-1ß and IL-18 but the mechanisms behind these processes are not fully understood. Here, the authors show that they can occur independently of the inflammasome adaptor ASC and without caspase-1 autoprocessing.

IL-10 engages macrophages to shift Th17 cytokine dependency and pathogenicity during T-cell-mediated colitis

The pro-inflammatory cytokine IL-1ß promotes the accumulation of pathologic Th17 lymphocytes that contribute to colitis. Here the authors show that the critical role of IL-10 in colitis suppression can be explained by its inhibition of IL-1ß production in macrophages.

NALP3 inflammasome upregulation and CASP1 cleavage of the glucocorticoid receptor cause glucocorticoid resistance in leukemia cells

William Evans and colleagues find that the proinflammatory genes CASP1 and NLRP3 are expressed at higher levels in primary acute lymphoblastic leukemia cells that are resistant than in those that are sensitive to glucocorticoid treatment. They find that CASP1 mediates glucocorticoid resistance through cleavage of the glucocorticoid receptor.

Pstpip2-mutant mice fed a high-fat diet are protected against inflammatory bone disease and bone erosion; this protection is associated with reductions in intestinal Prevotella levels and pro-IL-1β expression, and is dependent on the deletion of both caspases 1 and 8.
