Ketone α-alkylation at the more hindered site is challenging. Here, the authors report a highly regioselective nickel-catalysed allylic alkylation of unsymmetrical ketones at the more-hindered α-site with allylic alcohols.

Enantioselective synthesis of chiral amides by carbene insertion into amide N–H bond

Chiral amides are important structure in many natural products and pharmaceuticals, yet their efficient synthesis from simple amide feedstock remains challenge due to its weak Lewis basicity. Here, the authors report the enantioselective synthesis of chiral amides by N-alkylation of primary amides via Rh-catalyzed carbene N–H insertion reaction.

Achieving high enantioselective control in the hydrogenation of aliphatic ketones is a long-standing challenge. Now, the design of a spiro iridium catalyst with a narrow, crowded chiral pocket allows highly enantioselective hydrogenation of these compounds.

Stereospecific [1,3]-rearrangements are rarely reported method to efficiently build complex organic architectures. Here, the authors describe a metal-free intramolecular hydroalkoxylation/[1,3]-rearrangement sequence affording medium-sized lactams with wide scope, also in an asymmetric fashion.
