Colon cancer cells colonize the lung from established liver metastases through p38 MAPK signalling and PTHLH

Gomis and colleagues report on the mechanisms driving colon cancer metastasis. They show that whereas ERK2 activity promotes colon cancer metastasis to the liver, reduced p38 signalling upregulates the PTHLH cytokine to allow liver metastatic cell extravasation and colonization of the lung.

Long-term platinum-based drug accumulation in cancer-associated fibroblasts promotes colorectal cancer progression and resistance to therapy

Standard platinum-based chemotherapy is the basis of treatment of many cancers, however a proportion of patients do not derive benefit. Here the authors show that the platinum-based drug oxaliplatin accumulates in cancer-associated fibroblasts, activating pathways associated with cancer progression and resistance to therapy.

PD-L1 controls cancer pyroptosis

Programmed death ligand-1 (PD-L1) is well known for its role as an immune checkpoint regulator, but little is known about its function in other cellular processes. A study now shows that in tumour cells PD-L1 mediates pyroptosis, an inflammatory form of cell death, by activating the expression of Gasdermine C, ultimately leading to tumour necrosis.

Genes that mediate breast cancer metastasis to the brain

Little is known about the mechanisms by which breast cancer cells metastasize to the brain. By performing gene expression analysis on cells that preferentially infiltrate the brain it has now been possible to identify three genes that are involved in this process, two of which—COX2 and HBEGF—have previously been shown to mediate breast cancer metastasis to the lung.
