Loss of NUMB drives aggressive bladder cancer via a RHOA/ROCK/YAP signaling axis

The transition from non-muscle-invasive bladder cancer (NMIBC) to muscle-invasive bladder cancer (MIBC) is associated with poorer patient prognosis. Here, the authors investigate NUMB loss as a driver of the NMIBC-to-MIBC transition and identify a RHOA/ROCK/YAP signaling pathway as a therapeutic vulnerability.

CDK12 promotes tumorigenesis but induces vulnerability to therapies inhibiting folate one-carbon metabolism in breast cancer

Finding biomarkers for targeted therapy is a promising approach to treat cancer. Here, the authors show that in breast cancer preclinical models and patients, CDK12 promotes tumourigenesis but induces selective vulnerability to therapies that target folate one-carbon metabolism.

A self-sustaining endocytic-based loop promotes breast cancer plasticity leading to aggressiveness and pro-metastatic behavior

It is unclear if genetic alterations in endocytic proteins play a causal role in high incidence human cancers. Here, the authors report the oncogenic role of Epsin3 (EPN3) in breast cancer, and show EPN3 to drive tumorigenesis through induction of a partial epithelial mesenchymal transition state and a TGFβ-dependent regulatory loop that promotes cellular plasticity and invasive behaviour.

Antagonists of growth-hormone-releasing hormone: an emerging new therapy for cancer

Growth-hormone-releasing hormone (GHRH) and its receptors are expressed in a wide variety of normal and malignant tissues. This article describes various GHRH antagonists, their mechanisms of action, andin vivostudies. These antagonists are a promising potential therapy for a wide range of cancers, and perhaps other diseases.
