RNA catalyses nuclear pre-mRNA splicing

The spliceosome is shown to catalyse splicing through the RNA and not the protein components of the spliceosome; pre-messenger RNA splicing requires U6 snRNA acting by a mechanism similar to that used by group II self-splicing introns.

psiCLIP reveals dynamic RNA binding by DEAH-box helicases before and after exon ligation

ATP-dependent helicases remodel the spliceosome and proofread splice site recognition. A new method – Purified Spliceosome iCLIP (psiCLIP) – probes protein-RNA interactions in defined spliceosome complexes to reveal how the helicases Prp16 and Prp22 promote correct mRNA synthesis through dynamic binding on their RNA substrates.

De novo variants in the RNU4-2 snRNA cause a frequent neurodevelopmental syndrome

The non-coding RNA RNU4-2, which is highly expressed in the developing human brain, is identified as a syndromic neurodevelopmental disorder gene, and, using RNA sequencing, 5′ splice-site use is shown to be systematically disrupted in individuals with RNU4-2 variants.

Evidence for a group II intron–like catalytic triplex in the spliceosome

It has long been thought that the catalytic RNAs of self-splicing group II introns and the spliceosome function by similar mechanisms. Now, a combination of genetic, cross-linking, and biochemical analyses of yeast U6 snRNA provide compelling evidence that spliceosomal RNAs form triplex structures similar to those used by group II introns to catalyze splicing.

A conformational switch in PRP8 mediates metal ion coordination that promotes pre-mRNA exon ligation

The core of the spliceosome contains PRP8, a highly conserved protein that contacts each of the active site components and is implicated in both steps of pre-mRNA splicing. New work shows that structural rearrangement of the PRP8 RNase H domain between the first and second steps forms a Mg²⁺-binding site that is required for the second splicing reaction, revealing the molecular basis of the two-state model of splicing catalysis.
