Macrophages induce AKT/β-catenin-dependent Lgr5⁺ stem cell activation and hair follicle regeneration through TNF

Hair can be regenerated after skin wounding. Here the authors show that inflammatory macrophages produce TNF that activates Wnt signalling in hair follicle stem cells to drive this hair regeneration after wound repair in mice.

B-cell-derived lymphotoxin promotes castration-resistant prostate cancer

In a mouse model of prostate cancer it is shown that infiltrating B cells promote tumorigenesis by secreting lymphotoxin. Lymphotoxin accelerates the emergence of castration-resistant prostate tumours in this model. Interfering with this pathway may offer therapeutic strategies for androgen-independent prostate cancer.

Basophil-derived tumor necrosis factor can enhance survival in a sepsis model in mice

Basophils constitute <1% of peripheral blood leukocytes. Piliponsky and colleagues show that basophils serve as a critical component of antibacterial responses by secreting tumor necrosis factor at early time points after systemic infection.

Inflammation-induced formation of fat-associated lymphoid clusters

Fat-associated lymphoid clusters are lymphoid tissues that support B-1 cells. Caamaño and colleagues show that inflammation that elicits the cytokine TNF and activates natural killer cells contributes to the formation of these clusters in visceral fat.

Constitutive smooth muscle tumour necrosis factor regulates microvascular myogenic responsiveness and systemic blood pressure

TNF is typically viewed as an inflammatory mediator. Here the authors identify a non-inflammatory mechanism conserved across species whereby the constitutively expressed smooth muscle cell TNF mediates myogenic signal transduction in skeletal muscle resistance arteries and regulates mean arterial blood pressure.
