Fucosylated haptoglobin promotes inflammation via Mincle in sepsis: an observational study

Haptoglobin (Hp) scavenges cell-free haemoglobin and is associated with the prognosis of human sepsis. Here, the authors characterize the association of fucosylated Hp in sepsis progression and identify a macrophage population expressing a receptor for fucosylated Hp, highlighting its inflammation-amplifying effects in in vitro human cell cultures and in vivo using a mouse model.

A metazoan ortholog of SpoT hydrolyzes ppGpp and functions in starvation responses

SpoT has a key role in the bacterial starvation response. Now the metazoan ortholog of SpoT, Mesh1, has been identified, and the structure reveals a conserved active site that can catalyze ppGpp hydrolysis. The Drosophila Mesh1 deletion mutant has impaired starvation resistance, and microarray analysis gives further insight into the starvation response.

ETV4 is a mechanical transducer linking cell crowding dynamics to lineage specification

Yang, Golkaram et al. reported that in human embryonic stem cells, cellular crowding leads to the blockade of FGFR1 endocytosis, resulting in a decrease in ETV4 expression. This, in turn, derepresses the neuroectoderm fate.

Precise capture and dynamic relocation of nanoparticulate biomolecules through dielectrophoretic enhancement by vertical nanogap architectures

Label-free trapping of nanoparticles via dielectophoretic forces is traditionally done with electrodes in a horizontal gap layout. Here, the authors present a vertical nanogap architecture, which allows for precise capture and spatiotemporal manipulation of nanoparticles and molecular assemblies.

ALDH7A1 inhibits the intracellular transport pathways during hypoxia and starvation to promote cellular energy homeostasis

Intracellular vesicle transport can be regulated by Brefeldin‐A ADP‐Ribosylated Substrate (BARS) during vesicle fission. The late stage of COPI vesicle fission requires shorter forms of phosphatidic acid and diacylglycerol

Phosphatidic acid and diacylglycerol are required for COPI vesicle fission. Here, the authors find that shorter forms of these lipids promote the late stage of COPI vesicle fission, further suggesting how lipid geometry contributes to membrane deformation in driving fission.

Coordinated regulation of bidirectional COPI transport at the Golgi by CDC42

The COPI complex, which has a role in retrograde transport through the Golgi, is shown to also mediate anterograde tubular transport through the Golgi; in response to external stimuli, the small GTPase CDC42 acts as an essential modulator of bidirectional Golgi transport, and promotes the sorting of cargoes destined for anterograde transport into the tubules at the expense of those targeted for retrograde transport.
