Time-dependent catalytic activity in aging condensates

Kang et al. reveal that enzyme activity within biomolecular condensates diminishes with aging, as these liquid droplets transition to a solid state. This process leads to a significant decrease in their ability to enhance biochemical reactions.

Upregulation of WDR6 drives hepatic de novo lipogenesis in insulin resistance in mice

Yao and Gong et al. identify WD40 repeat-containing protein 6 (WDR6) to be upregulated in the liver of insulin-resistant mice. WDR6 contributes to promoting hepatic de novo lipogenesis during insulin resistance by upregulation of fatty acid synthase, and the authors identify a small molecule to inhibit this effect of WDR6 and reduce hepatic steatosis.

A truncating variant of 17β-hydroxysteroid dehydrogenase-13 (17β-HSD13), a lipid droplet -associated protein, associates with lower risk of chronic liver disease. Here the authors identify a phosphorylation site in 17β-HSD13 which promotes lipolysis, and report that a knock-in mouse with a 17β-HSD13 mutant defective for phosphorylation is more susceptible to nonalcoholic steatohepatitis.

Development of nanodrug-based eye drops with good penetration properties and ROS responsiveness for controllable release to treat fungal keratitis

We developed a ROS-responsive and controllable nanocarrier (GC-EB) that efficiently delivered a clinically used antifungal drug, voriconazole (VOR). GC-EB-VOR exhibited high penetration through corneal barriers, good retention in the cornea and controllable drug release under low concentrations of ROS. Mechanistically, the successful delivery and accumulation of GC-EB-VOR in the cornea followed by the inhibition of oxidative stress and of the inflammatory response contributed to the therapeutic effect of GC-EB-VOR against fungal keratitis.
