IGF1 drives Wnt-induced joint damage and is a potential therapeutic target for osteoarthritis

Osteoarthritis is a common, yet untreatable, joint disease associated with high Wnt signaling, but how Wnt causes joint destruction is unknown. Here, the authors show that Wnt induces IGF1 which leads to joint damage, and targeting this mechanism offers a potential therapeutic opportunity in osteoarthritis.

DOT1L safeguards cartilage homeostasis and protects against osteoarthritis

DOT1L is one of the few genes linked to osteoarthritis by human GWAS. Here the authors show that DOT1L-dependent histone methylation protects homeostasis of articular chondrocytes by SIRT1-dependent inhibition of canonical WNT signalling, and that inhibition of DOT1L can drive osteoarthritic disease in mice.

A Notch in the joint that exacerbates osteoarthritis

Tight regulation of signalling cascades is vital for the correct development and function of bones and joints. A new study suggests that Notch signalling might join the likes of the transforming growth factor superfamily and Wnt signalling cascades as having an important function in joint homeostasis and disease.

Identification of renin progenitors in the mouse bone marrow that give rise to B-cell leukaemia

Renin cells have traditionally been associated with the kidney where they regulate blood pressure and fluid electrolyte homeostasis. In this study, Belyea et al.describe a renin progenitor in the bone marrow that gives rise to B-cell leukaemia when RBP-J, the final effector of the Notch pathway, is deleted.

In this Review, the role of canonical Wnt signalling in articular cartilage is discussed, along with the regulatory mechanisms that exist to fine-tune Wnt signalling and the rationale for developing drugs that modulate Wnt signalling for the treatment of joint diseases such as osteoarthritis.
