  • Fezf2 (Fezl) is a transcription factor that specifies corticospinal motor neurons (CSMN) originating from cortical layer 5b. Lodato et al. use cortical progenitor isolation from developing mouse brain and gene expression profiling to identify genes downstream of Fezf2 and demonstrate co-regulation of CSMN gene ensembles by Fezf2 in establishing CSMN cell identity.

  • TALEs (transcription activator–like effectors) contain a large number of nearly identical repeats, which makes it difficult to synthesize new variants. Feng et al. describe a facile method for assembling TALEs and show TALEs' utility for activating expression of endogenous human genes.

  • The mechanisms underlying neuron specification and maturation are unclear. Here the authors provide an integrated epigenomic and transcriptomic analysis of mouse and marmoset neocortical neuronal classes. Pan-neuronal programs active during early development are more evolutionary conserved but not neuron-specific, whereas pan-neuronal programs active during later stages of maturation are more neuron- and species-specific.
