The AKT2/SIRT5/TFEB pathway as a potential therapeutic target in non-neovascular AMD

Lysosomes maintain RPE health via TFEB/E3-regulated autophagy. Here, the authors show deregulated AKT2/SIRT5/TFEB signaling in the RPE inhibits both lysosomal and mitochondrial function and leads to AMD, suggesting this pathway might provide a therapeutic target for AMD.

Reducing Akt2 in retinal pigment epithelial cells causes a compensatory increase in Akt1 and attenuates diabetic retinopathy

Retinal pigment epithelium (RPE) plays an important role in diabetic retinopathy (DR) progression. Here, the authors show AKT1 and AKT2 are reciprocally regulated in the RPE of DR donor tissue and diabetic mice, reducing Akt2 in RPE causing a compensatory increase in Akt1 and attenuating DR.

Impaired endolysosomal function disrupts Notch signalling in optic nerve astrocytes

Crystallins are structural proteins that are expressed on the outside of the lens of the eye. Valapala and colleagues find that specific crystallins in retinal astrocytes regulate V-ATPase activity and endolysosomal acidification, to facilitate optimal Notch signalling during retinal development.

βA1-crystallin regulates glucose metabolism and mitochondrial function in mouse retinal astrocytes by modulating PTP1B activity

Ghosh et al. show that a lens protein βA1-crystallin inhibits PTP1B activity, regulating the metabolism of retinal astrocytes. They find that knockdown of βA1-crystallin exacerbates the streptozotocin-induced diabetic retinopathy-like phenotype in mice, suggesting the possibility that the interaction between βA1-PTP1B may be targeted for diabetic retinopathy.

βA3/A1-crystallin regulates apical polarity and EGFR endocytosis in retinal pigmented epithelial cells

Shang et al. utilize a conditional Cryba1 knockout mouse model to demonstrate a key role for βA3/A1-crystallin in retinal pigmented epithelium. As a result of βA3/A1-crystallin deficiency, the authors report EGFR endocytosis defects and actin network disruption which leads to polarity disruption and degeneration through PITP β/PLC signaling.

Neutrophils homing into the retina trigger pathology in early age-related macular degeneration

Ghosh et al. find that in a mouse model, age-related macular degeneration (AMD) is associated with retinal infiltration of neutrophils, activation of interferon lambda and production of other cytokines. They also see increased interferon lambda production in the eyes of AMD patients.
