Analyses of 475 ancient horse genomes show modern horses emerged around 2200 bce, coinciding with sudden expansion across Eurasia, refuting the narrative of large horse herds accompanying earlier migrations of steppe peoples across Europe.

Stabilizing a metalloid {Zn₁₂} unit within a polymetallide environment in [K₂Zn₂₀Bi₁₆]⁶⁻

Low-valent zinc clusters, though exceedingly rare, are appealing synthetic targets because there is evidence that they may show unconventional chemical and physical behavior. Here, the authors obtain a large heterometallic zinc-bismuth cluster anion and discover that it bears a metalloid {Zn₁₂} core with four-center bonding and essentially zero-valent character.

Substantial π-aromaticity in the anionic heavy-metal cluster [Th@Bi₁₂]⁴⁻

A wide variety of organic and inorganic compounds show π-aromaticity, yet for all-metal systems it has remained restricted to compounds with three to five atoms. Now, the anionic cluster [Th@Bi₁₂]⁴⁻ has been shown to exhibit π-aromaticity, with a significant ring current despite relying on the delocalization of only two π-electrons.

Effective engineering of a ketoreductase for the biocatalytic synthesis of an ipatasertib precursor

Ipatasertib is a potent Akt (protein kinase B) inhibitor synthesized via a chemoenzymatic process. Here, the authors use mutational scanning and algorithm-aided enzyme engineering to optimize a ketoreductase from Sporidiobolus salmonicolor and generate a 10-amino acid substituted variant exhibiting a 64-fold higher k_cat and improved yield for the relevant alcohol intermediate, with ≥ 98% conversion and a diastereomeric excess of 99.7% (R,R-trans) from 100 g L⁻¹ ketone after 30 h.
