Rewiring macrophages for anti-tumour immunity

Tumour-associated macrophages facilitate cancer progression, but whether they can be reprogrammed to elicit an anti-tumour response remains unclear. Deletion of the microRNA-processing enzyme Dicer is now shown to rewire macrophages to an anti-tumour mode, leading to an enhanced response to immunotherapy and inhibition of tumour progression.

Single cell analysis in head and neck cancer reveals potential immune evasion mechanisms during early metastasis

The molecular mechanisms underlying lymph-node metastasis in head and neck squamous-cell carcinoma remain to be investigated. Here, the authors perform single-cell RNA sequencing of cancer cells and CD8 + T cells and suggest potential mechanisms of immune evasion during early metastasis.

The macrophage tetraspan MS4A4A enhances dectin-1-dependent NK cell–mediated resistance to metastasis

The membrane tetraspan protein MS4A4A is expressed on tissue-resident and tumor-associated macrophages. Locati and colleagues show that MS4A4A colocalizes with the β-glucan receptor dectin-1 to enhance cytokine and reactive oxygen species production and to enhance NK cell–mediated control of tumor metastasis.

WIP1 phosphatase is a negative regulator of NF-κB signalling

NF-κB activity is positively regulated through phosphorylation, but how activation is reversed has been unclear. A genome-wide siRNA screen reveals that the phosphatase WIP1 dephosphorylates the NF-κB subunit p65 at Ser 536, a phospho-residue critical for transcriptional regulation.
