Single-cell analysis identifies the CNP/GC-B/cGMP axis as marker and regulator of modulated VSMCs in atherosclerosis

The second messenger 3’,5’-cyclic guanosine monophosphate (cGMP) is important for the maintenance of cardiovascular homeostasis. Here, correlative single-cell profiling of cGMP dynamics and vascular cell phenotype reveals a druggable atheroprotective mechanism and suggests parallels between remodeling processes in bone and atherosclerotic arteries.

Cre/lox-assisted non-invasive in vivo tracking of specific cell populations by positron emission tomography

Non-invasive cell tracking is a powerful method to visualize cells in vivo under physiological and pathophysiological conditions. Here Thunemann et al. generate a mouse model for in vivo tracking and quantification of specific cell types by combining a PET reporter gene with Cre-dependent activation that can be exploited for any cell population for which a Cre mouse line is available.

A shear-dependent NO-cGMP-cGKI cascade in platelets acts as an auto-regulatory brake of thrombosis

Nitric oxide (NO) inhibits thrombosis in part by stimulating cyclic guanosine monophosphate (cGMP) production and cGMP-dependent protein kinase I (cGKI) activity in platelets. Here, Wen et al. develop a cGMP sensor mouse to follow cGMP dynamics in platelets, and find that shear stress activates NO-cGMP-cGKI signaling during platelet aggregation to limit thrombosis.
